Antivirulence activities of retinoic acids against Staphylococcus aureus

چکیده
Multidrug-resistant bacteria such as Staphylococcus aureus constitute a global health problem. Gram-positive S. secretes various toxins associated with its pathogenesis, and biofilm formation plays an important role in antibiotic tolerance virulence. Hence, we investigated if the metabolites of vitamin A 1 might diminish toxin production. Of three retinoic acids examined, 13- cis -retinoic acid at 10 μg/mL significantly decreased without affecting planktonic cell growth (MIC >400 μg/mL) also inhibited by epidermidis μg/mL), but less affected uropathogenic Escherichia coli strain, Vibrio or fungal Candida strain. Notably, all- trans hemolytic activity staphyloxanthin production . Furthermore, transcriptional analysis disclosed that repressed expressions virulence- biofilm-related genes, two-component arlRS system, α-hemolysin hla , nuclease ( nuc1 nuc2 ), psmα (phenol soluble modulins α) In addition, plant nematode toxicity assays showed was only mildly toxic concentrations many folds higher than effective antibiofilm concentrations. These findings suggest particularly acid, be useful for suppressing virulence characteristics

Discovery of antivirulence agents against methicillin-resistant Staphylococcus aureus.
Antivirulence agents inhibit the production of disease-causing virulence factors but are neither bacteriostatic nor bactericidal. Antivirulence agents against methicillin-resistant Staphylococcus aureus (MRSA) strain USA300, the most widespread community-associated MRSA strain in the United States, were discovered by virtual screening against the response regulator AgrA, which acts as a transcr...
متن کاملComparison of Efficacy of Two Disinfectants Against Foodborne Staphylococcus aureus
Background and Objectives: Foodborne illness is a common public health problem. Surfaces and equipment used in food production are involved in the spread of foodborne pathogens. The aim of this study was to evaluate and compare the antimicrobial effect of Vinoxide and Sanisept against Staphylococcus aureus strains isolated from food production lines. Material and Methods: In this descriptive s...
متن کاملVaccination against Staphylococcus aureus pneumonia.
BACKGROUND Staphylococcus aureus causes serious infections in both hospital and community settings. Attempts have been made to prevent human infection through vaccination against bacterial cell-surface antigens; thus far all have failed. Here we show that superantigens and cytolysins, when used in vaccine cocktails, provide protection from S. aureus USA100-USA400 intrapulmonary challenge. MET...
متن کاملTeichoic acids in pathogenic Staphylococcus aureus.
Twenty-six strains of Staphylococcus aureus obtained from patients with endocarditis were studied for the production of alpha- and/or beta-ribitol teichoic acid (TA), using highly specific anti-TA antibodies prepared in rabbits. A counterimmunoelectrophoretic assay was used. Beta-TA was the predominant residue produced by all strains; alpha-TA was found in all strains, but in smaller amounts an...
متن کاملAntibacterial mechanism of fraxetin against Staphylococcus aureus
Fraxetin is one of the main constituents of the traditional medicinal plant Fraxinus rhynchophylla. The inhibitory effect of fraxetin on various bacterial strains has been extensively reported, however, its mechanism of action on bacterial cells remains to be elucidated. In the present study, the antibacterial mechanism of fraxetin on Staphylococcus aureus was systematically investigated by exa...
